Biosynthesis of Nicotinamide Mononucleotide: Current Metabolic Engineering Strategies, Challenges, and Prospects

Nicotinamide mononucleotide (NMN) is an essential precursor of nicotinamide adenine dinucleotide (NAD+), which widely applied in the pharmaceutical and biotech industries. The biosynthesis NMN currently attracting much attention because it has non-toxic reaction conditions low amounts isomers, whereas chemical synthesis yields not environmentally friendly. This review systematically describes two biosynthetic pathways detail for first time introduces latest studies on production through different using metabolic engineering strategies. accumulation can be improved by optimizing activity key enzymes, enhancing supply precursors co-factors, inhibiting byproducts, promoting product export. Finally, we also discuss current challenges producing possible solutions future.
